Following M. tuberculosis infection, C57BL/6J.CC058 Tip5 congenic mice exhibit increased splenic bacterial burden relative to matched wild-type siblings. This increase was significant in pilot studies after adjustment for sex (p = 0.04). This observed phenotype is consistent with the original genetic mapping of Tip5. In the Collaborative Cross study, Tip5 was associated with both spleen chromosome equivalents (CEQ) and spleen colony-forming units (CFU) following M. tuberculosis infection. The NOD founder haplotype at this locus was associated with higher values for splenic bacterial burden.
The Tip5 interval was also associated with IL-10 abundance. The QTL for IL-10 overlapped the Tip5 bacterial burden interval, and the NOD haplotype was associated with high spleen CFU, spleen CEQ, and IL-10 values. Coincident mapping of these traits supports a possible single causal variant with pleiotropic effects.

A genomic region containing Tip5 from the CC058/Unc strain was introduced onto the C57BL/6J background through congenic backcrossing. Selection during strain development was guided by MiniMUGA array genotyping. The strain underwent six generations of backcrossing when it was genotyped and transitioned to random intra-strain mating.
MiniMUGA genotyping performed in mid-2024 defined the retained CC058-derived interval as approximately 170.69–182 Mb on chromosome 2. Eight diagnostic markers distinguish C57BL/6J and CC058 within this interval. In the genetic mapping study, Tip5 was mapped to distal chromosome 2 and was associated with splenic bacterial burden following Mycobacterium tuberculosis infection. Spleen chromosomal equivalents (CEQ) mapped to a 174.29–178.25 Mb interval with a peak at 178.25 Mb, while the associated spleen CFU interval extended from 73.98–180.10 Mb.

Distribution of this strain requires submission of the MMRRC Conditions of Use (COU). A link to the COU web form will be provided via email after an order has been placed; the form should be completed then or the email forwarded to your institutional official for completion.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.
2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.
3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
