Genetic Alterations
CRISPR-targeting inserted a CAG-LSL-cAMPFIRE conditional overexpression cassette into the locus. The cAMP Fluorescence Imaging Reporters based on Epac (cAMPFIREs) is composed of a modified RAPGEF3 (M312L, L777A, K778A), mTurquoise2, and Venus (Addgene 182284). Cre-mediated recombination removed the floxed stop cassette to allow for expression of the reporter.

Phenotype
Mice globally expressing cAMPFIRE-M do not exhibit overt abnormalities and are phenotypically normal under baseline conditions. Homozygous mice are viable and fertile, with no reported morphological or behavioral abnormalities.

Following Cre-mediated recombination, the reporter is constitutively expressed. Mice exhibit visible fluorescence, allowing genotype identification by observation with fluorescence-detecting goggles.

Strain Development
The cAMPFIRE-M reporter line was generated by CRISPR/Cas9-mediated insertion of a conditional reporter cassette into the Rosa26 locus. The donor vector was microinjected into one-cell embryos isolated from C57BL/6J mice. Injected embryos were transfered into CD1 pseudo-pregnant foster females. Founders were initially screened for the presence of the expression cassette and then confirmed by PCR. Confirmed positive founders were bred with wildtype animals and positive F1 pups were further confirmed for the expression cassette. Mice were then bred with Cre-expressing recombinase in the oocyte (129S1/Sv-Hprt1tm1(CAG-cre)Mnn/J) that was bred to C57BL/6J. The globally expressing cAMPFIRE-M mice were maintained as homozygous breeding pairs.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
