Genetic Alterations
The Apob allele codes for "VHLTPVEKSAVT" and "KEFTPPVQAAYQ" instead of "LSVKAQYKKNSD" and "GTSRLMRKRGLK" of the wild-type (apoB100) allele at residues 3143 to 3154 and 3356 to 3366, respectively, in exon 26 of the Apob gene. The two domains of apolipoprotein B are predicted to facilitate the binding and clearance of LDL particles through the LDL receptor (LDLR). Replaced amino acid sequences are from the human beta-globin. The precise replacement of approximately 30 bp of each DNA was achieved by "in" and "out" two-step recombination resulting in duplication and deletion of the targeted locus. There are no extra DNA fragments remaining in the mouse genome such as selectable marker genes and loxP sequences.

    Phenotype
    Homozygous: ApoB100beta homozygotes have slightly reduced plasma lipoprotein levels and are protected from diet-induced hyperlipidemia. On Ldlr-/-, Apobec1-/- background, homozygotes do not accumulate LDL and are protected from atherosclerosis.

    Heterozygous: Heterozygotes have an intermediate phenotype.

    Strain Development

    A mutation in the Apob gene was made in E14 cells (129P2/OlaHsd). Mice made from the modified cells were backcrossed towards C57BL/6J >8 generations. Subsequently, the mutants were crossed with Ldlr-/- mice (made with AB1/129S7 ESCs) on a C57BL/6J background and with Apobec1-/- mice on 129SvEv (gift from Dr. Eddy Rubin at UC-Berkley).

    A sample from this MMRRC strain was analyzed using the Mouse Universal Genotyping Array (MUGA) and MMRRC computational tools were used to assess the genetic background. A summary of the data can be found here.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
