Genetic Alterations
A LacZ gene-trap cassette was knocked into the endogenous Ube3a locus via homologous recombination in C57BL6 mouse ES cells. This allele is known as a "knockout First allele ", as it is initially non-expressive, but can be converted to a conditional allele via Flp recombination. Germline chimeric males (determined by the transmission of coat color in parallel breeding) were then crossed to C57BL/6 female homozygous Rosa26-FLPe mice (B6.129S4-Gt(ROSA)26Sortm1(FLP1)Dym/RainJ, Jackson Stock #009086) in order to excise the FRT-flanked lacZ gene trap from the Ube3aKO1st allele and thereby produce the Ube3aFLOX allele.

    Phenotype

    Homozygous: unknown

    Hetero/Hemizygous: Mice are viable and fertile and are obtained at mendelian ratios regardless of parental inheritance of the allele. When the inherited allele is maternally inherited, Ube3a protein expression in the brain is drastically reduced. Cellular, physiological, and behavioral phenotypes have yet to be explored.

    Strain Development
    C57BL/6N germline male chimeras were bred to C57BL/6J wild type females, resulting in heterozygous Ube3a knockout first females, which were in turn bred to a wild type C57BL/6J wild type male. One of the heterozygous male offspring was mated to a C57BL/6J wild type female to produce the heterozygous male mice to be donated.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
