Phenotype
Homozygotes express 10% of the wild-type CD45 levels; heterozygotes express 5%. Evidence suggests the full-length, non-mutated CD45 protein of strain LP4 is reduced due to low transgene copy number. The transgene has the added feature of being flanked by LoxP sequences, such than crosses of LP4 mice with cre-expressing strains result in transgene excision and 0% CD45 expression in cells expressing cre. The transgene encodes the CD45.1 allelic sequence which is detected by anti-CD45.1 antibodies. The strain exhibits some reduction in litter frequency, but lifespan and general health are otherwise normal.

Strain Development
The CD45-minigene transgenic construct 'VGI-1084' was injected into C57BL/6J zygotes, and 7 founders were identified: LP4, LP5H, LP5L, LP8, LP10, LP26, and LP28. VGI-1084 contains the same CD45 minigene construct as VGI-0020 (also known as pLFAiT200 in PMID:12393487) but has a LoxP site added at each end of the minigene. The LoxP sites allow the minigene to be deleted from the genome in the presence of cre. CD45-minigene transgenic LP4 (the ‘LP’ series designating the presence of the LoxP sites) was one of the founders. The LP4 founder was mated to C57BL/6J mice and progeny were screened for the presence of the transgene. The colony was then crossed onto the CD45-null background by matings with CD45 knockout mice also on the C57BL/6 background (JAX Strain #002725). LP4 mice homozygous for the transgene locus and on the CD45 knockout background were then sibling mated for at least 10 generations to date.

CD45 minigene strain LP4 is one of a series of transgenic strains prepared with the CD45 minigene and expresses the lowest amount of CD45 available, 10% of the wild-type when homozygous for the transgene and 5% as heterozygous. This level is lower than that of the lightning strain with a mutated CD45 sequence (JAX Strain #016091) and lower than that of the CD45-minigene transgenic strain B (MMRRC:44051). The transgene is flanked by LoxP sites and the cross of strain LP4 with vav-icre mice (JAX Strain #008610) results in offspring with 0% expression of CD45 in hematopoietic cells. Strain LP4 mice are therefore suitable for generating conditional mutants lacking CD45 expression in specific hematopoietic subpopulations using the many strains available with cell type-specific cre expression. The strain should be similar to the CD45-minigene transgenic strain B for studies on the effect of CD45 expression level, and thus low CD45 phosphatase activity, on lymphocyte activation and maturation. This strain should be also suitable for similar activation and maturation studies of other hematopoietic lineages which express CD45, especially using the LoxP feature with conditional cre expression. Due to the properties of the minigene, the normal cell lineage-specific isoform regulation is maintained in the minigene transgenics.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
