Genetic Alterations
Exon 9 was replaced with a loxP site flanked neomycin resistance gene cassette. The absence of transcript expression was confirmed by RT-PCR on olfactory mucosa, liver, lung, kidney, and testis extracts.

Phenotype
Cyp2a5-null mice have a lower hepatic nicotine 5′-hydroxylation activity in vitro, and slower systemic clearance of both nicotine and cotinine in vivo.

Strain Development
The targeting vector was prepared in a pMC-lox-neo-lox vector and consists of a tk-neo-bpA cassette for the neomycin-resistance (neo) gene expression, using the thymidine kinase (tk) promoter; the neo was flanked by two loxP sites. A 1.6-kb PstI fragment encompassing exons 6 to 8 and a 5.6-kb BamHI-EcoRI fragment of the 3′-flanking region were cloned into the pMC-lox-neo-lox vector. The vector was linearized and electroporated into Bruce4 (B6.Cg-Thy1a) ES cells. ES cells from positive clones were karyotyped and injected into albino B6(Cg)-Tyrc-2J/J blastocysts. Blastocysts were then transferred into the uterus of a pseudopregnant B6CBAF1/J mouse. Adult chimeras were bred with C57BL/6J female mice to produce germline-transmission F1 mice that were heterozygous for the Cyp2a5-null allele. F2 homozygotes were used for breeding and characterization

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
