Genetic Alterations
This strain harbors a Rln1-Uhrf2 duplication. Two loxP sites flanking the Rln1-Uhrf2 chromosomal segment were introduced into the germline, one via gene targeting in ES cells and the other by CRISPR/Cas-9 in zygotes. Mice with these two loxP sites in trans and the Hprt-Cre transgene were bred with wild type mice. Offspring containing an allele that had undergone trans-allelic recombination, resulting in a Rln1-Uhrf2 duplication, were identified.

Phenotype
This strain contains a homozygous duplication of the Rln1-Uhrf2 genomic segment (4 copies instead of the normal 2 copies). This model was created to to fine-map the observed molecular and behavioral phenotypes observed in the mice with the Rln1-Gldc duplication (MMRRC: 72077). Behavioral tests indicate that Rln1-Uhrf2 segment does not contribute to the phenotypes of the mice with the Rln1-Gldc duplication (MMRRC: 72077).

Strain Development
Two loxP sites flanking the Rln1-Uhrf2 genomic segment were inserted into the mouse genome, one loxP site via gene targeting in ES cells, the other one by CRISPR/Cas-9 in C57BL/6 zygotes. Mice carrying both loxP sites and an Hprt-Cre transgene were generated and bred with C57BL/6J wild type mice. Offspring were screened for the presence of the Rln1-Uhrf2 duplication. In case the cre transgene was present, it was bred out. Mice were bred to C57BL/6J wild type mice for at least 8 generations. Mice were then inbred for 5 generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
