A 23 bp deletion in exon 1 of the Ism1 gene was generated using CRISPR-Cas9 genome editing. This deletion causes an out-of-frame mutation that results in a disruption of the ISM1 protein and complete loss of functional (whole body knockout) ISM1 expression throughout the body.

Strain Description

Phenotype
Ism1 knockout (Ism1-/-) mice are viable, fertile, and do not exhibit any gross developmental or behavioral abnormalities.

Under baseline conditions, Ism1-/- mice develop spontaneous and progressive postnatal emphysema, with a milder phenotype on the C57BL/6J background than the FVB/NTac background. The emphysema phenotype becomes evident by 8 weeks of age and includes alveolar enlargement, extracellular matrix deterioration with elastin fiber loss and septal rupture, increased alveolar macrophage accumulation, elevated MMP-12 and MMP-9 expression, chronic low-grade inflammation, and mucus hypersecretion. Pulmonary testing at 2 months demonstrates increased total lung capacity, elevated residual volume, increased static and dynamic compliance, reduced FEV100, and increased airway resistance, consistent with hyperinflation and airflow obstruction seen in human chronic obstructive pulmonary disease (COPD).

In addition to spontaneous emphysema, Ism1-/- mice exhibit exaggerated inflammatory responses to inducible lung injury models. These include heightened airway inflammation in LPS-induced acute lung injury, inceased airway hyperresponsiveness in a house dust mite-induced asthma-like model, and enhanced lung inflammation and pulmonary fibrosis in the bleomycin-induced pulmonary fibrosis model.

The C57BL/6J Ism1-/- strain represents a model of impaired lung homeostasis with susceptibility to chronic inflammation, emphysema, airway hyperreactivity, and fibrosis.

Strain Development
Ism1 knockout mice were generated via CRISPR/Cas9-mediated genome editing on two backgrounds, FVB/NTac and C57BL/6J mice. This strain (RRID:MMRRC_076033-UCD) was developed on the C57BL/6J background.

Briefly, CRISPR guide(s) and Cas9 protein were microinjected into C57BL/6J zygotes and resulting progeny were screened for the desired mutation by PCR and sequencing. Founder mice were bred to C57BL/6J to establish the line through the N1 and N2 generations. N2 heterozygous mice were intercrossed to generate N3 homozygous phenotyping cohorts, which were then mated to obtain more homozygous mice. Prior to being accepted into the MMRRC, the line was maintained on the C57BL/6J background by periodic outcrossing of N3 homozygous mice to wild-type C57BL/6J every 6–12 months to produce heterozygous mice for colony maintenance.
