TBR1 is a potential "master regulator" of autism risk by virtue of its direct regulation of an autism-associated gene network in human midfetal brain (Willsey et al., 2013; Notwell et al., 2016).

Tbr1em2Boro (Tbr1K228E) show normal growth and body/brain mass, and normal motor coordination (negative geotaxis) during early postnatal time points.

Tbr1K228E mice exhibit elevated Tbr1 protein levels due to transcriptional upregulation:
  • Heterozygous mice:
    • show 2x Tbr1 protein levels compared to wild-type mice
    • lack the posterior limb of the anterior commissure (AC)
  • Homozygous mice:
    • show 5x the Tbr1 protein levels compared to wild-type mice
    • show ectopic expression of Tbr1 protein across all cortical layers
    • show more severe defects—absence of AC, misdirected callosal fibers, abnormal organization of internal capsule axons, perinatal lethality, and small olfactory bulbs
    • do not show a significant increase on cortical apoptosis (unlike KO mice)

Strain Development
CRISPR synthetic guide RNA (sgRNA) design generated Tbr1em2Boro (Tbr1K228E) using the CRISPOR tool. C57BL/6NJ zygotes were coinjected with Cas9 protein and single-stranded-oligodeoxynucleotide (ssODN) donor containing the patient mutation (confirmed via sequencing screening). Embryos were transplanted into pseudopregnant recipient female CD-1 mice, and founders from these litters were first mated to C57BL/6NJ breeders (JAX Stock #005304). Then, derived N1 progeny, (first identified via sequencing), were mated again to C57BL/6NJ, generating N2 mice (also identified via sequencing). N2 hets were mated for production of initial phenotyping cohorts. Tbr1K228E mice were genotyped using PCR amplification of the mutation-containing genomic region, followed by restriction enzyme digest of the PCR product.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
