Genetic Alterations
This allele carries a targeted mutation in the Mgrn1 gene, introducing an arginine-to-glutamic acid substitution at codon 318 (p.R318E) through two nucleotide changes (c.1238C>G and c.1239G>A). The donating investigator reports that this knock-in completely disrupts MGRN1 E3 ubiqutin ligase activity without disrupting its interaction with known substrates (based on cell culture data, ubiquitination assays).

Phenotype

The R318E mutation disrupts the allosteric linchpin residue within the RING domain of Mgrn1 that is essential for engaging ubiquitin and its E2 conjugating enzyme through hydrogen bonding. As a result, Mgrn1R318E is catalytically inactive and unable to mediate substrate ubiquitination. In the presence of transmembrane adapters MEGF8 or ATRNL1, catalytically inactive MGRN1R318E does not promote melanocortin receptor (MCR) ubiquitination and degradation. Thus, while the mutation abolishes E3 ubiquitin ligase activity, it preserves normal binding interactions between MGRN1 and its known substrates.

Homozygous Mgrn1R318E mice develop congenital heart defects and die before or shortly after birth.

Strain Development
Full-length mouse Mgrn1 containing a C-terminal 3xFLAG tag was synthesized as a gBlock and used as a template for generating all Mgrn1 constructs. The R318E mutation was introduced by overlap extension PCR.

Briefly, Mgrn1 constructs were first cloned into the pENTR2B plasmid and then into the pEF5/FRT/V5-DEST expression vector. A CRISPR/Cas9 ribonucleoprotein (RNP) complex and a single-stranded oligodeoxynucleotide (ssODN) were electroporated into one-cell C57BL/6J embryos to introduce the R318E substitution. Founders were identified by sequencing across the target region followed by ICE analysis. Verified founders were crossed to wildtype C57BL/6J mice, and after three generations of backcrossing, heterozygotes were intercrossed.

Colony and Husbandry Information

Breeding has been a combination of backcross (to WT C57BL/6J) and intercross of heterozygous siblings. Line can be maintained by mating heterozygotes to wildtype C57BL/6J or by intercrossing heterozygotes. All breeding performance data were obtained from heterozygous mice, as homozygous mutants are nonviable and do not survive beyond birth.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients.
