Strain Development
A Chd8 floxed allele was generated using CRISPR/Cas9 endonuclease-mediated genome editing. Guide RNAs (Chd8_int2_sgRNA1: attgattgcccagtagctca, Chd8_int2_sgRNA2: ccagcttgaccttgagctac, Chd8_int3_sgRNA1: GACTGCTCTCTGCTGCCCCG and Chd8_int3_sgRNA2: GCTATCTCCAGCACCCCACG) were selected to introduce loxP sites flanking exon 3 of the chromodomain helicase DNA binding protein 8 (Chd8) on chromosome 14. Chd8 transcript Chd8-201 (ENSMUST00000089752.10) was used as reference for exon number and sequence. The gRNAs, double-stranded donor plasmid and Cas9 nuclease were introduced to single cell C57BL/6J zygotes and transferred to pseudo pregnant females. Correctly targeted pups were identified by sequencing and PCR and were bred for at least 2 generations after which, the Chd8 floxed mice were bred to Cre recombinase expressing mice, B6.Cg-Edil3Tg(Sox2-cre)1Amc/J resulting in excision of exon 3. The resulting Chd8 knock-out (KO) mice were maintained on a C57BL/6J background by the donating laboratory.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
