Genetic Alterations
Itgal (LFA-1) knock-out mice exhibit defective leukocyte adhesion, reduced NK cytoxicity, absent DTH, and a selective defect in the induction of peripheral immune responses. LFA-1 exons 2, 3, 4, 5 and part of exon 6 are disrupted with a Neo insertion in reverse orientation. These mice may be useful for studies of lymphocyte activation and proliferation.

Phenotype
The targeted Itgal (LFA-1) gene encodes the integrin alpha L chain and is involved in leukocyte intercellular adhesion and lymphocyte costimulator signaling. It is essential for cell-mediated cytotoxicity by cytotoxic T- lymphocytes (CTL) and natural killer (NK) cells. Mice homozygous for the knockout allele are viable and fertile, exhibiting no overt phenotype. Leukocytes from LFA-1 deficient mice fail to aggregate in response to PMA, indicating a failure of homotypic leukocyte adhesion. In addition, T cells exhibit a reduced proliferative capacity in mixed lymphocyte reaction (MLR) assays, NK cytotoxicity is reduced and delayed-type hypersensitivity (DTH) is absent. Although mice can mount a normal CTL response to LCMV and VSV infections, they fail to prime or reject injected tumor cells. These mice may be useful for studies of lymphocyte activation and proliferation.

Strain Development
A targeting vector containing a neomycin cassette, in reverse orientation, was used to disrupt exons 2, 3, 4, 5, and part of exon 6. Exons 1-6 contain the signal peptide and the amino terminal extracellular region. The construct was electroporated into 129P2/OlaHsd derived E14K embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. The resulting chimeric animals were bred to C57BL/6J to achieve germline transmission. The mice were then backcrossed to C57BL/6 for at least 8 generations. To establish a live colony, an aliquot of frozen sperm is used to fertilize C57BL/6J.
