Strain Development
  1. Targeting vector based on pKO scrambler V915 (Lexicon Genetics Inc., Woodland, TX) flanking a LoxP-pGK-Neo-LoxP selection cassette containing a BAC clone insertion containing exons 6-9 of murine gene Pten (Gene ID 19211, NM_008960.2) with variant c.719A>T, predicting p.Tyr240Phe (Y240F).
  2. Electroporation of linearized target vector into TC1 ES cells (origin: 129S6/SvEvTac)
  3. G418 selection, clonal expansion, and screening via Southern for correct targeting into the Pten locus
  4. Two ES clones injected into blastocysts; implantation into C57BL/6 female to produce chimeras
  5. Tail snip PCR to confirm genotype of chimeras
  6. Mating of chimera offspring (>80% chimerism based on agouti coat) with C57BL/ for germline transmission
  7. Southern blot screening to ascertain germline transmission
  8. Heterozygous mice bred with E2a-cre (Jackson lab, #003314) mice to excise the pGK-Neo selection cassette; progeny screened by PCR to confirm cre-mediated excision.
  9. Heterozygous mice lacking pGK-Neo were bred with B6.129S2-Trp53tm1Tyj/J mice (Jackson Lab #002101) to generate Y240FKI/KI; P53-/- mice.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
