Genotype Determination
Phenotype
The mahogany allele reduces the pheomelanic phenotype and the obesity phenotype in mice that are yellow and obese because of mutation at the agouti locus. Mahogany mice also are more active than normal mice, and they have lesions of the brain that increase with age. Black mahogany mice are difficult to differentiate by phenotype from black wild type mice. Therefore, the Ay allele was used for backcrossing mahogany onto C57BL/6J using C57BL/6J-Ay/a mice. The Ay allele is maintained in the stock to differentiate between mahogany and wild-type mice, and also to study the phenotypic and genic interactions between the genic functions of the attracting locus and the agouti locus.

Colony and Husbandry Information

This strain has mutations in two genes. The mahogany allele of the attractin gene is homozygous in this strain (Atrnmg/Atrnmg). The yellow allele of the agouti gene (Ay) is also present in 50% of the offspring (this allele is lethal in the homozygous state so animals are heterozygous for the yellow allele and the nonagouti allele (Ay/a)). Animals heterozygous for the yellow allele & homozygous for the mahogany allele (Ay/a, Atrnmg/Atrnmg) will have a dark brown mahogany coat color and have poor reproduction but allow visualization of the mahogany allele. Animals that do not carry the yellow allele (a/a, Atrnmg/Atrnmg) are black and have good breeding performance but the mahogany coat color is masked by the black coloration of the haircoat.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
