Genetic Alterations
The targeting construct was designed and generated by the Sanger Institute and ES cells were obtained from the UC Davis KOMP Repository (ES line JM8.N4 strain C57BL/6N). It is a Knockout-first, conditional ready, LacZ-tagged vector to target the Sned1 locus, GenBank file here.

Phenotype
Most knockout mice die within 2 days of being born. The rare knockout survivors present severe cranio-facial defects (shorter snouts, asymmetrical nasal cavities) and growth retardation. Heterozygous mice are normal.
Strain Development
Two targeted Sned1tm1a(KOMP)Wtsi ES cell clones (EPD0300_5_02 E02 and F01) from the JM8.N4 parental ES cell line generated by the Wellcome Trust Sanger Institute were obtained from the KOMP Repository at UC Davis and cultured on feeder cells using the recommended media including the 2i additives MEK inhibitor PD0325901 and GSK3 inhibitor CHIR99021 (StemGent). Three subclones of each clone were sent to Cell Line Genetics (Madison, WI) for cytogenetic analysis and two sub-clones of the targeted clone E02 were found to have an apparently normal mouse male karyotype. Cells from sub-clone E02-2D2 were microinjected into BALB/c blastocysts obtained from natural matings as previously described - 10 recipient females each received 14 embryos. Seven pregnant females gave birth to 41 pups, of which 6 showed at least 70% dark coat color against white, BALB/c fur, and allowed establishment of the Sned1tm1a(KOMP)Wtsi (referred to as Sned1LacZ-Neo) mouse line.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
