This line is a conditional-ready Retnla strain, where Cre-expression will lead to inversion of a tdTomato reporter gene, deleting Retnla exons 2-4. The neomycin selection cassette of the original allele was removed through Flp recombination.

Strain Development

A reporter marker cassette was inserted in antisense orientation downstream of Retnla exon 4, such that cre-mediated deletion of the targeted exon 2 to exon 4 will flip the reporter gene (tdTomato) into sense orientation leading to its expression. To enhance the stability of the reporter messenger and to increase the chance of having a bright signal, a WPRE sequence (woodchuck hepatitis virus post-transcriptional regulatory element - see PMID:12787924) was added to the construct.

Vector design: Previously isolated homology fragments and cDNA were used to generate the targeting vector (NAI1-HR targeting vector for Retnla Exons 2-4) for homologous recombination in electroporated ES cells derived from C57BL/6N (genOway, proprietary). Following neomycin selection and clonal expansion, Southern blotting confirmed correct targeting. Six ES clones were injected into blastocysts and implanted into pseudopregnant dams. Tail-snip PCR confirmed the genotype of the chimeras, whose offspring were mated with C57BL/6N (Charles River) for germline transmission. Three founders were identified and bred with C57BL/6J (RRID:IMSR_JAX:000664) mice for two generations after the selection cassette had been removed with a C57BL/6N Flp recombinase strain (genOway, proprietary).

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
