Genetic Alterations
Sycp2l is located on Chr 13 and consists of 21 exons. In the targeted allele, deletion of exons 8-14 (encoding residues 191-383) leads to a frameshift in the transcript.Homozygous female Sycp2l knockout mice exhibit a progressive loss of oocytes and decreased fertility. These mice may be useful in the study of female reproductive aging.

Phenotype
SYCP2L (synaptonemal complex protein 2-like) is a sequence homolog of SYCP2 and is expressed only in oocytes. Variants in human SYCP2L are associated with Age at Natural Menopause (ANM). SYCP2L localizes to the centromere of dictyate oocytes. The Sycp2l knockout allele has a loxP-flanked PGK neo cassette replacing exons 8-14. Homozygous female mice undergo a progressive reduction of ooctyes with aging; by 14 months oocyte numbers drop to 20% of controls. Primordial follicle number decreases beginning at 2 months of age; decreases in numbers of primary and secondary follicles are apparent at 12 months of age. Although homozygous female mice produce the same number of litters as controls, litter size is significantly decreased. Male fertility is unaffected. These mice may be useful in the study of female reproductive aging.

Strain Development
A targeting vector was designed by Dr. Jeremy Wang (University of Pennsylvania) to replace exons 8-14 of the synaptonemal complex protein 2-like gene (Sycp2l) on chromosome 13 with a loxP-flanked PGK-neo cassette leading to a frameshift in the transcript. The construct was electroporated into (C57BL/6 x 129S4/SvJae)F1-derived V6.5 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts, and chimeric males were bred with C57BL/6J females to establish the colony. The resulting mice were bred with C57BL/6 wildtype mice for one generation prior to sending males to The Jackson Laboratory Repository MMRRC in 2015. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
