    Errfi1 genomic DNA was isolated from R1 ES cells. To generate the targeting vector, the Errfi1 fragment with exon 2-4, 2-kb fragment of the 5' arm and 2-kb fragment of the 3' arm were subcloned into the TOPO-cloning vector (Invitrogen, Carlsbad, CA) using PCR reactions. High-fidelity Taq polymerase was used (Roche Applied Science, Indianapolis, IN) for PCR amplification, and all of the PCR products were confirmed by sequencing. One LoxP site followed by the PGK-neo cassette (phospho-glycerol kinase neomycin-resistance flanked with two FRT sites) was inserted downstream of exon 4. The other LoxP site was inserted upstream of exon 2. The 5' ends of the targeting vector were constructed by cloning 2kb of Errfi1 homologous sequence upstream of Mig-6 exon 2, whereas the 3' end contains a 2-kb homologous sequence downstream of Errfi1 exon 4. The linearized targeting construct was transfected into R1 ES cells by electroporation. Neomycin-resistant clones were screened by Southern-blot analysis using 5' external probes with EcoRI digested genomic DNA. Four of 192 clones were targeted. Correctly targeted clones were microinjected into blastocysts derived from C57BL/6 mice. Seven chimeras were bred to C57BL/6 mice, and F1 agouti offspring mice were analyzed by PCR genotyping to validate the germ line transmission of the Errfi1 floxneo allele (PMID:1798766). The neo cassette was removed with C57BL/6-Tg(ACTFLPe)9205Dym (MGI allele #2448985) strain obtained from the animal facility at Baylor College of Medicine, Houston TX.
