Genetic Alterations
A major extracellular adenosine-producing enzyme, ecto-5'-nucleotidase, is expressed by gene Nt5e and was targeted for a frameshift mutation. A targeting vector replaced exon 2 of the Nt5e gene (flanked by loxP sites) with a neo-cassette, flanked by
FRT sites resulting in a deletion/knock-out of Nt5e.

    Heterozygous and homozygous: normal enzymatic activity of NT5E (CD73).

    Conditional knock-out allele phenotype: Floxed Nt5e mice, when crossed with Albumin-Cre expressing mice, result in loss of CD73 from the liver. This deletion of Nt5e yields a 70% decrease in total CD73 protein, thus preventing CD73's major enzymatic role in dephosphorylating extracellular adenosine monophosphate (AMP) into anti-inflammatory adenosine leading to spontaneous age- and sex-dependent liver disease development.

    Strain Development
    Nt5e floxed (CD73fl/fl) mice were generated by Cyagen Bioscience, Inc. Exon 2 of the CD73-encoding gene, Nt5e was targeted to generate a frameshift mutation. The template for the targeting vector was a bacterial artificial chromosome (BAC) clone RP23-137M4 from the C57BL/6J library. The targeting vector consisted of the following: a Neo cassette flanked by FRT sites and exon 2 in gene Nt5e was flanked by loxP sites.

    Linearized targeting vector was electroporated into unspecified ES cells, followed by neomycin selection of resistant clones. Targeted ES clones were then injected into blastocysts and transferred into surrogate mothers to obtain chimera lines. The conditional knockout allele was obtained after flippase-mediated recombination. Heterozygous mice (CD73fl/+) were bred to each other to obtain homozygous mice (CD73fl/fl). For the deposited strain, CD73fl/fl mice were backcrossed to C57BL/6J WT mice >10 times.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
